Application

 

Preparation method of 3,5-difluoro-2,6-diaminopyridine:
(1) Add 2,3,4,5,6-pentachloropyridine, solvent, metal reducing agent and ammonium chloride into the reactor according to the amount of metal reducing agent being 0.5~1.5 equivalents, the reaction temperature is 10~100° ℃, the reaction time is 1 to 4 hours; reduction is performed to obtain 2.3.5.6-tetrachloropyridine, the metal reducing agent is any one of aluminum powder, iron powder, tin particles and zinc particles or the solvent is methanol, ethanol, dihydrogen Any one or combination of methyl chloride, toluene, chloroform and acetonitrile.

(2)The ion exchange reaction is carried out to obtain 2.3,5,6-tetrafluoropyridine by adding 2.3,5,6-tetrachloropyridine, catalyst, fluorinating agent, and polar aprotic solvent to the reactor in accordance with the amount of fluorinating agent being 4~8 equivalents. The reaction temperature is 120~260°C, and the reaction time is 6 to 18 hours. The catalyst is chosen from among tetrabutylammonium chloride, tetrabutylammonium fluoride sulfone, benzyltriethyl ammonium vapor (TEBA), tetrabutylammonium bromide (TBAB), tetrabutylammonium bisulfate, trioctylmethylammonium chloride, dodecyltrimethylammonium chloride, and tetradecyltrimethylammonium chloride. The polar aprotic solvent is any one or multiple combinations of N,N-dimethylformamide, N,N-dimethylacetamide, dimethyl sulfoxide, and sulfolane. one or more ammonium chloride combinations;
(3) Fill the reactor with 2.3.5.6-tetrafluoropyridine and ammonia water in a 1:2 molar ratio. Toluene solution is the solvent. 20 to 90°C is the reaction temperature. Two to six hours is the reaction time. The process of aminolysis 3.5-difluoro-2.6-diaminopyridine should be obtained.
