Application

 

Step 1. Add 50 grams of tap water and 14 grams of 25% ammonia (15 ml, 0.206 mol) into a 250 ml three-necked bottle. Cool to 10°C and slowly add 15.9 g (0.100 mol) o-fluorobenzoyl chloride dropwise. The reaction will be obviously exothermic. The jacket is cooled to control the temperature to 0-5°C. After the dropwise addition is completed, the reaction is maintained at 10-15°C for 4 hours, filtered with suction, and the filter cake is washed with 15 grams of tap water and dried at 70-75°C at normal pressure to obtain o-fluorobenzyl. Amide 12.5 g. Yield 90%

Step 2: Get a thermometer, condenser tube, and addition funnel ready in a 500 ml three-necked flask that has been cleaned and dried. Add 116.9g of toluene (135ml), 27g of o-fluorobenzamide (0.194mol), and 60.0g of phosphorus oxychloride (0.39mol) gradually and dropwise while keeping the temperature below 60°C. Continue raising the temperature to 90–100°C once the dropwise addition is finished, then allow the reaction to continue for an hour, cool somewhat, then evaporate the toluene under reduced pressure until the temperature does not surpass 100°C and the vacuum does not fall below -0.08MPa. To bring the pH down to 7-8, add 43.4 grams (50 milliliters) of toluene after cooling, and then gradually add 10% sodium hydroxide aqueous solution dropwise. To eliminate toluene, separate the phases, evaporate the organic phase at lower pressure, and then distill under reduced pressure to obtain 2-fluorobenzonitrile. 18.8 grams, yield 80%.
